摘要
目的:观察通脉化浊汤对实验性动脉粥样硬化(AS)大鼠血清中载脂蛋白A-I(apoA-I)、载脂蛋白B-100(apoB-100)含量的影响,探讨通脉化浊汤抗AS作用和机理。方法:144只SD大鼠,按体重将大鼠随机分为6组,即:正常组、模型组、通脉化浊汤低剂量组、通脉化浊汤中剂量组、通脉化浊汤高剂量组、西药对照组。除正常组饲以普通饲料外,其它各组给予维生素D370万IU/kg3天内灌胃,而正常组给予相同体积生理盐水灌胃,饲以高脂饲料,建立动脉粥样硬化模型大鼠,各组分别在治疗第7、14、28天3个时间点取材,采用快速免疫比浊法,检测apoA-I、apoB-100水平。结果:与模型组相比,通脉化浊汤各剂量组apoB-100水平均显著降低,同时apoA-I水平显著提高;与西药对照组相比,通脉化浊汤组与其无显著差异。结论:通脉化浊汤可降低AS大鼠apoB-100水平,提高apoA-I水平,通过调节载脂蛋白的水平,纠正脂质代谢紊乱,发挥抗AS作用。
Objective :To observe the effect of Tongmai Huazhuo ( TMHZ ) decoction on the level of apolipoprotein A-I ( apoA-I ) and apolipoprotein B-100 ( apoB-100 ) in the serum of atherosclerosis ( AS ) rats, and discuss its effect and mechanism to resist AS. Method : 144 SD rats were randomized into six groups : normal group, model group, low-dose TMHZ decoction group, middle-dose TMHZ decoction group, highdose TMHZ decoction group and control group of Western medicine. Rats from the normal group were fed with standard chow and lavaged with normal saline, while rats from the rest groups were lavaged with vitamin D 3.7 × 10^6IU/kg for 3 days and fed with high fat chow to induce AS models of rats. After 7,14 and 28 days, at each time point rats were sacrificed to take sample. Then the levels of apoA-I and apoB-100 were tested by ITM Immunoturbidimetry. Result:There was no statistical significance between model group and TMHZ decoction group. Conclusion :TMHZ decoction can reduce AS rats' level of apoB-100, while increase their level of apoA-I, and through regulating the level of apolipoprotein, it corrects lipid metabolic disturbance, resisting AS.
