摘要
目的分析宫颈癌患者放射治疗过程中病灶局部肿瘤浸润淋巴细胞亚群的变化。方法收集8例接受局部放射治疗的宫颈癌患者放疗前、放疗第1周、第2周、第3周的肿瘤组织标本,采用免疫组织化学方法原位分析CD8^+、Granzyme B^+、OX40^+、FOXP3^+、CD4^+浸润T淋巴细胞状态的动态变化。结果放射治疗后,CD8^+、Granzyme B^+、CD4’T淋巴细胞在肿瘤实质的分布均有下降,且差异均有统计学意义(PCD8+=0.023〈0.05、PGra+:0.015〈0.05、PCD4+=0.022〈0.05);OX40^+、FOXP3^+T淋巴细胞在肿瘤实质的分布虽略呈下降之势,但差异均无统计学意义(P〉0.05)。肿瘤间质的CD8^+、Granzyme B^+、FOXP3^+、CD4^+T淋巴细胞在接受放射治疗后分布虽有变化,但差异均无统计学意义(P〉0.05)。OX40’T淋巴细胞接受放射治疗后在肿瘤间质的分布略有升高,且差异有统计学意义(POX40+=0.039〈0.05)。结论局部放疗对宫颈癌局部细胞毒性淋巴细胞有明显抑制作用.而调节性T细胞表现出较强的放射抵抗性。
Objective To analyze the dynamic changes of tumor infiltrating lymphocyte subsets in cervical cancer site during the course of local radiotherapy. Methods Eight cases of cervical cancer were recruited, and the tumor tissue specimens were taken before radiotherapy, at first week, second week and third week of radiotherapy, respectively. CD8^+ , Granzyme B^+ , OX40^+ , FOXP3^+ , and CD4^+ phenotypes were used to probe the tumor infiltrating lymphocyte subsets by immunohistochemistry. Results After radiotherapy the distribution of CD8^+ , Granzyme B ^+ , CD4 ^+ T lymphocytes in tumor solid components were decreased, and the differences were statistically significant (PCD+ =0.023 〈0.05, PGra+ =0.015 〈 0.05, PCD4 + = 0. 022 〈 0.05 ). But OX40^+ and FOXP3^+ T cells showed slightly decrease, and there was no significant difference ( P 〉 0. 05 ). CD8^+ , Granzyme B^+ , FOXP3^+ and CD4 + T cells in tumor stroma didn' t show significant change after radiotherapy ( P 〉 0.05 ). OX40 ^+ T cells in tumor stroma increased and the difference was statistically significant (Pox40^+ = 0. 039 〈 0.05 ). Conclusion The cytotoxic T cells are more sensitive but regulatory T cells are more resistant during local radiotherapy.
