摘要
目的观察金纳多联合钙离子阻滞剂对冠心病大鼠药酒中毒心肌细胞凋亡的影响。方法将构建成功的80只冠心病大鼠药酒中毒模型分为4组,即模型组、金纳多治疗组、尼莫地平治疗组、金纳多联合尼莫地平组。根据不同组别行药物治疗,分别于治疗后不同时间点取材。采用免疫组化方法检测心肌组织中Bc1-2、Caspase-3蛋白的表达,以末端脱氧核糖核素转移酶介导的duTP缺味端标记法检测大鼠心肌凋亡细胞。结果模型组大鼠Bc1-2降低而Caspase-3表达增高,金纳多治疗组、尼莫地平治疗组大鼠Bcl-2表达增高、Caspase-3表达降低,与模型组对比差异有统计学意义(P<0.05);金纳多联合尼莫地平组Bcl-2表达明显增高、Caspase-3表达明显降低,与金纳多治疗组、尼莫地平治疗组比较差异有统计学意义(P<0.01)。结论金纳多联合钙离子阻滞剂可显著提高冠心病大鼠药酒中毒Bcl-2蛋白的表达、抑制Caspase-3蛋白表达、降低凋亡细胞数,从而抑制心肌细胞凋亡。提示金纳多联合钙离子阻滞剂可能通过抑制心肌细胞的凋亡而治疗冠心病大鼠药酒中毒。
Objective To observe the effect of Ginaton combined calcium inhibitor on apoptosis of myocardial cell of coronary heart disease in rats after liquor poisoning. Methods 80 rats with coronary heart disease after liquor poisoning were divided into four groups: Each group divided into four time points. According to different group injected different drug and got samples after treatment at different time. The expression of the Bcl - 2 and Caspase - 3 were examined by immunohistochemistry techniques and the apoptosis of myocardial cell were examined by TUNEL method. Results In the model group, levels of Bcl -2 expression decreased, but levels of Caspase -3 expression increased. Comparing with model group, levels of Bcl -2 expression increased, levels of Caspase -3 expression decreased in Ginaton treatment group and Nimodipine treatment group ( P 〈 0.05 ) ; Comparing with Ginaton treatment and Nimodipine treatment, levels of Bcl - 2 expression obviously increased, levels of Caspase - 3 expression obviously decreased in Ginaton combined Nimodipine treatment ( P 〈 0.01 ). Conclusion Ginaton combined calcium inhibitor can significantly increase levels of Bcl - 2 of rats with coronary heart disease after liquor poisoning, suppress levels of Caspase - 3, decrease cell population of apoptosis, suppress of ap- optosis myocardial cell. Ginaton combined calcium inhibitor treat coronary heart disease in rats after liquor may through suppress of apoptosis myocardial cell.
