摘要
在pH为7.4的Tris-HCl缓冲溶液中,应用紫外光谱法和荧光光谱法对文多灵与鲱鱼精DNA分子间的相互作用进行了研究.研究结果表明,DNA与文多灵存在较强的相互作用,计算了25℃和40℃两个不同温度下文多灵与DNA的结合常数和结合位点数分别为5.26×103L/mol、1.0411和7.77×103L/mol、1.056.紫外光谱实验结果表明,DNA的加入可使文多灵252 nm处的紫外吸收峰发生显著升高和蓝移;荧光探针实验表明文多灵不能使小檗碱-DNA体系的荧光发生猝灭,证明文多灵是以沟槽结合的方式与DNA结合.
The interaction of Vindoline and DNA was studied by UV spectrometry and fluorescence spectrometry in tris-HCl buffer solution of pH 7.4.. The results showed that Vindoline can react with DNA. The binding constant K and the binding sites number n at 25 ℃ and 40 ℃ were calculated as 5.26×10^3 L/mol, 1.041 land 7.77×10^3 L/mol, 1.056 respectively. The UV spectrometry result showed that the addition of DNA can cause the significant blue shift of the absorbance peak and increase of absorbance; the fluorescent probe test showed that Vindoline can not cause the fluorescence quenching of BR-DN A system, which proved that ;indoline binds with DNA in a mode of groove binding.
