摘要
[目的]探讨PPARγ-2基因多态性与广西巴马小型猪2型糖尿病易感性的关系。[方法]利用高糖高脂日粮饲喂24头广西巴马小型猪,利用PCR方法扩增PPARγ-2基因外显子2部分序列,测定其空腹血糖和胰岛素(INS)含量并进行糖耐量试验。[结果]克隆的PPARγ-2基因外显子2部分序列存在1处SNP位点(19813A/G),有AG(7头)和AA(17头)2种基因型。AG型广西巴马小型猪的空腹胰岛素含量和糖耐量试验中60 min血糖值显著高于AA型(P<0.05),而AG型广西巴马小型猪2型糖尿病发生率(71.4%)明显高于AA型(5.9%)。[结论]广西巴马小型猪PPARγ-2基因外显子2的19813A/G多态性与2型糖尿病易感性有关。
[Objective] The research aimed to discuss the relationship between the genetic polymorphism of PPARγ-2 and the susceptibility of type 2 diabetes mellitus(T2DM)in Guangxi Bama mini-pig.[Method] 24 Guangxi Bama mini-pigs were fed with high-fat and high-sucrose diet and partial sequences of exon 2 of PPARγ-2 gene were amplified by PCR method.The contents of blood glucose and insulin in Guangxi Bama mini-pig under fasting conditions were determined and the glucose tolerance test(GTT) was also carried out.[Result] There was one SNP site(19813A→G)in partial sequence of exon 2 of the cloned PPARγ-2 gene.And two genotypes,AA(7 pigs) and AG(17 pigs)were detected.The fasting blood insulin content and 60 min blood glucose level in GTT in AG-genotype Guangxi Bama mini-pig were significantly higher than that of AA genotype(P0.05).The incidence of T2DM in AG-genotype Guangxi Bama mini-pig(71.4%)was obviously higher than that of AA genotype(5.9%).[Conclusion] The polymorphism of 19813A→G in exon 2 of PPARγ-2 gene was related with the susceptibility of T2DM in Guangxi Bama mini-pig.
