肝癌细胞FDG摄取与葡萄糖转运蛋白相关性的实验研究 被引量:2

摘要 目的观察高低转移潜能肝癌细胞摄取FDG的特性,及其与葡萄糖转运蛋白表达的相关性。方法两株不同转移潜能的肝癌细胞与FDG行细胞结合实验,观察不同转移潜能细胞对FDG摄取能力的差异。两株细胞与FDG共同孵育60 min,洗脱后不同时间点测定细胞内示踪剂的吸收率和细胞外液内的流出率。Western blot法检测Glut1和Glut3的表达,并与细胞FDG摄取率进行相关分析。结果高、低转移潜能肝癌细胞对FDG摄取率差异无统计学意义(P>0.05)。高、低不同转移潜能细胞流出率、吸收率差异有统计学意义(P<0.05),流出率和吸收率存在着明显的正相关关系(P<0.05)。Glut1和Glut3在两株细胞之间差异无统计学意义,与细胞FDG摄取率存在着不同的相关性。结论 FDG摄取率不能有效地区分肝癌的高低转移特性。肝癌细胞株FDG吸收率与流出率的特性是高低转移特性的具体体现。葡萄糖转运蛋白的表达与FDG摄取情况存在着不同的相关性,进一步验证了肝癌高低转移特性的差异。 Objective To observe the characteristics of high and low metastatic potential HCC cells uptake FDG, and the correlation of it and glucose transporter expression. Methods FDG-cells binding experiments were carried out. FDG uptake pattern was observed and the different metastatic potential HCC cells were analyzed. The cells and FDG were co-incubated 60 minutes, and the outflow and absorption rates were measured each 20 min interval. Expression of Glutl and Glut3 were detected in two cells line. Results Between the high and low metastatic potential of hepatocellu- lar carcinoma cells, FDG uptake rate was no statistical difference (P 〉 O. 05). But outflow and absorption rates had sta- tistical difference between two cells line (P 〈 O. 05). Outflow and absorption rates had significantly positive correlation by Pearson correlation test. FDG uptake related indices Glutl and Glut3 were no statistical difference between two cells line. FDG uptake rates with Glutl and Glut3 of two cells line existed the different correlation. Conclusion FDG uptake rate can' t distinct the different potential of HCC cells. FDG outflow and absorption rates in HCC cells line with different metastatic potential are the concrete performance of hepatocellular carcinoma biological behavior.
