苦参碱联合顺铂对肺腺癌A549细胞表达的影响 被引量:4

摘要 目的:观察苦参碱联合顺铂对人肺腺癌A549细胞抑制效应,探讨作用机制。方法:进行人肺腺癌A549细胞培养,噻唑蓝法检测不同浓度苦参碱、顺铂单药及联合用药组对人肺腺癌A549细胞增殖影响。用逆转录聚合酶链反应(RT-PCR)法分别检测苦参碱及联合用药组中血管内皮生长因子mRNA的表达。结果:苦参碱、顺铂单药抑制A549细胞的生长并呈时间、剂量依赖性(P<0.05)。与顺铂单药组相比,联合用药组能明显抑制肺腺癌A549细胞的生长(P<0.05)。顺铂单药及与不同剂量的苦参碱联合组能抑制VEGF mRNA的表达,与对照组比较VEGF mRNA的表达显著下降(P<0.05)。且发现所观察的苦参碱联合顺铂组与顺铂单药组相比明显抑制VEGF mRNA的表达(P<0.05)。结论:苦参碱、顺铂单药可浓度、时间依赖性抑制A549细胞的生长。苦参碱可增强顺铂对A549细胞的细胞毒性,该作用与抑制细胞内血管内皮生长因子mRNA的表达有关。推测通过抑制血管内皮生长因子的表达,苦参碱和顺铂在抑制肺癌血管的生长机制中可能具有一定作用。 Objective:To explore the effect of Matrine combined with cisplatin on proliferation of A549 lung ade- nocarcinoma cells cultured in vitro,and explore its mechanism. Methods:A549 celline was treated with different concen- tration of Matrine and cisplatin,individually or in combination.And then cellular proliferation was evaluated by MTT method . VEGF expression in A549 cells in different concentrations of cisplatin and Matrine combined with cisplatin group was detected by RT-PCR analysis.Results:MTT assay showed that Matrine and cisplatin could also inhibit the pro- liferation of A549 cells in a dose and time-dependent manner in vitro (P〈0.05) .Compared with Matrine and cisplatin indi- vidual drug groups ,Matrine combined with cisplatin groups enhanced the growth inhibition in A549 cells.And the ex- pression of VEGF mRNA was downregulated significantly in A549 ceils treated by different concentrations of Matrine combined with cisplatin groups (P〈0.01) in a dose-dependent way. Conciusion:Matrine and cisplatin could inhibit the proliferation of A549 cell line in a dose and time-dependent manner, respectively. Matrine could increase the cytotoxic ef- fect of cisplatin on A549 cell .It can be inferred that Matrine and cisplatin may inhibit lung cancer vascular growth at- tributable in part to downregulation expression of VEGF.
