摘要
对克隆获得的转录因子NF-κB/Rel家族成员Relish基因在斑节对虾发育阶段的表达特征进行分析。所获的PmRelish是Relish基因的亚型之一,与凡纳滨对虾LvRelish高度相似,结构域组成相同,其中RHD、IPT和DD结构域均具有100%的同源性,ANK结构域的相似性也高达97.7%。NF-κB/Rel家族进化树分析显示,PmRelish与对虾、昆虫等节肢动物的Relish聚为一支,并与P105(NF-κB1)和P100(NF-κB2)最为相近。PmRelish的表达在4个幼体发育时期都存在显著性差异,其中糠虾阶段表达量最高。PmRelish在不同发育阶段的精巢和卵巢中的表达特征相似,即未成熟期的表达相对较低,其中PmRelish在10~12 cm体长雄虾精巢中表达量最高,卵巢中的表达则在成熟期(V期)最高,为卵原细胞期的14.3倍。
The paper obtained a transcription factor Relish from Penaeus monodon, a member of the NF-KB/Rel family, and analyzed its expression profile at the different developmental stages. PmRelish, a subtype of the Relish gene, is highly similar to LvRelish from Litopenaeus vannamei, with the same composition of domains. Their RHD, IPT and DD domain have 100% homology, and the similarity of the ANK domain is up to 97.7%. Phylogenetic tree of NF-KB/Rel family shows that PmRelish is clustered with other arthropods Relish including other shrimps and insects, and near to P105 (NF-KB1) and P100 (NF-KB2). The expression of PmRelish among four periods of larval development has a significant difference, with the highest expression level in mysis stage. The expression profile of PmRelish at different developmental stages in testis is similar to that in ovary, with a low expression level at immature phases. The expression level of PmRelish is highest in testis of male tiger shrimp with body length of 10-12 cm, and is highest in mature stage ovary of female tiger shrimp with a 14.3 folds higher than that in oogonium stage.
