木质层孔菌诱导漆酶及部分酶学性质研究 被引量:1

摘要 采用单因素试验的方法,对木质层孔菌产漆酶的培养条件进行优化,并研究了漆酶的性质。结果表明,在以1 mmol/L愈创木酚为诱导剂的条件下,漆酶培养的最佳碳源是小麦秸,最佳氮源是牛肉膏,最适培养温度是28℃,最佳培养时间是192 h;漆酶在反应温度32℃、pH 4.0、底物浓度1.2 mmol/L的环境条件下酶活性最高,金属离子对其活性也有一定的影响,其中Cu2+对漆酶有激活作用,Fe2+和Mn2+对漆酶活力有一定的抑制作用,Mg2+和Na+对漆酶影响不大。 In this research,used enzyme activity as the evaluation index,through the optimization of enzyme production conditions,we screened out the optimal culture method of Fomes lignosus during laccase production,and studied the partial enzyme action characteristics.The results showed that the optimal culture method of Fomes lignosus during laccase production was as follows,in the condition of using 1 mmol/L guaiacol as inducer,the best carbon source was wheatstraw,the best nitrogen source was beef paste,the optimum culture temperature was 28℃,the optimum culture time was 192 h.The optimum pH of laccase was 4,the optimum substrate concentration was 1.2 mmol/L,the optimal reaction temperature was 32℃.Laccase could be activated by metal ion Cu2+,Fe2+ and Mn2+ had certain inhibitory action on the laccase activity,Mg2+ and Na+ had little effect on laccase activity.
