过氧化氢抑制PC12细胞IGF-1促存活功能的体外研究 被引量:1

摘要 目的研究过氧化氢(H2O2)对PC12细胞胰岛素样生长因子(IGF-1)促存活功能的影响及可能机制。方法不同浓度的H2O2预处理PC12细胞60 min,MTT及Western blot方法观察H2O2对IGF-1促存活及其受体酪氨酸磷酸化的影响;不同的抑制剂预处理研究H2O2影响IGF-1促存活功能的可能机制。结果 H2O2预处理可以剂量依赖性的降低PC12细胞IGF-1的促存活功能及其受体的磷酸化水平,NMDA受体的特异性拮抗剂MK801可以逆转H2O2的抑制作用。结论 H2O2可以通过谷氨酸受体亚型NMDA受体抑制IGF-1R的磷酸化,从而抑制其促存活作用。 Objective To investigate the influence and underlying mechanisms of hydrogen peroxide ( H2O2 ) on the survival promoting effect of insulin-like growth factor 1 (IGF-1) in PC12 cells. Methods Pretreatment with different con- centrations of H2O2 in PC12 cells for 60 minutes, MTF assay and Western blot were performed to examine H2O2 in influen- cing the survival promoting effect of IGF-1 and the tyrosine phosphorylation of IGF-1 receptor. We investigated the underly- ing mechanisms by using different antagonists. Results Pretreatment of H2O2 could attenuate survival promoting effect of IGF-1 and its receptor tyrosine phosphorylation in a dose-dependent manner, while MK801 could reserve the inhibitory effect of H2O2. Conclusion H2O2 inhibits IGF-1 receptor tyrosine phosphorylation and the survival promoting effect by NMDA receptor.
