摘要
目的研究黄芪的最佳提取工艺。方法以黄芪总皂苷和黄芪甲苷的提取率为指标,分别用紫外分光光度法和薄层扫描法进行测定,用正交试验法优选黄芪提取工艺。结果加水量、提取时间对水提黄芪总皂苷提取率有显著性差异(P<0.05),而提取次数无显著性差异(P>0.05);对黄芪甲苷水提提取率,3个因素都有显著性差异(P<0.05)。醇提取时间、乙醇浓度对黄芪总皂苷及黄芪甲苷提取率均有显著性差异(P<0.05),而提取次数均无显著性差异(P>0.05)。结论黄芪的最佳提取工艺为16倍量水提取4次,每次1 h。
Objective To investigate the best extraction technique of radix astragali. Methods The extraction rates of total saponins of astragalus and astragaloside IV were used as the index and were detected respectively by ultraviolet speetrophotometry and TLC scanning. The optimizing extraction technique of radix astragali was selected by orthogonal experiment. Results When water was used as the solvent,the amount of added water and extracting time were factors that significantly affected the extraction of total saponins of astragalus ( P 〈 0.05 ) ,while the times of extraction did not affect the extraction( P 〉 0.05 ). These three factors significantly affected the extraction of astragaloside IV ( P 〈 0. 05 ). When ethanol was used as the solvent in the extraction of total saponins of astragalus, the extracting time and the concentration of alcohol significantly affected the extraction,while the times of extraction did not affected that(P 〉 0.05). Conclusion The best method of radix astragali abstraction is using 16 times of water to extract 4 times and for 1 h at each time.
