摘要
目的:观察以中药传统方剂生肌散为基础的改良新药创愈生肌散对糖尿病大鼠模型慢性难愈创面的治疗作用。方法:对糖尿病大鼠模型慢性难愈创面经随机分组后给予外用创愈生肌散和rb-bFGF,以未作处理的创面作为空白对照。分别在伤后第1、4、7、10、14和21天观察各组慢性难愈创面的愈合情况,使用RealtimePCR方法检测各时间点慢性难愈创面中VEGF、HIF-1α以及Angl基因的表达情况。结果:在糖尿病I陛慢性难愈创面模型中,创愈生肌散组糖尿病性慢性难愈创面的愈合时间为(21.3±2.1)天,rb-bFGF组创面的愈合时间为(23.2±2.4)天,对照组创面的愈合时间为(26.8±2.8)天,差别有统计学意义(P〈0.05)。使用RealtimePCR方法检测发现,创愈生肌散组的创面标本中的VEGF、HIF-1α以及Angl表达量明显高于其他两组,差别有统计学意义(P〈0.05)。结论:使用创愈生肌散可明显缩短糖尿病I陛慢性难愈创面的愈合时间,创愈生肌散可通过上调VEGF、HIF-1α以及Angl等与新生血管密切相关的基因表达而促进创面中的血管生成。
Objective To investigate the therapeutical effect of Chuangyu Shengji Powder on chronic wound of diabetes rat model, which is a new clrug based on traditional prescriptions. Methods The present study utilize diabetes animal model on rat to form chronic wound to dectect the therapeutical effect and potential mechanism of Chuang Yu Sheng Ji Powder. Compare the wound healing time of animal model cured by Chuangyu Shengji Powder with that of blank control. Investigate the expression of VEGF, HIF-1α and Angl, which are closely related to angiopoiesis, in tissue sample of chronic wound by Real time PCR. Results The wound healing time of animals with diabetes treated with Chuangyu Shengji Powder, Recombinant Bovine Basic Fibroblast Growth Factor and blank control were (21.3±2.1)d, (23.2±2.4)d and (26.8±2.8)d respectively,among which the difference was statistically significant (P〈0.05). Real time PCR showed that the expression of VEGF, HIF-1α and Angl were up-regulated in animals treated with Chuangyu Shengji Powder, compared with animals treated with Recombinant Bovine Basic Fibroblast Growth Factor and blank control (P〈0.05). Conclusion The results above showed that Chuangyu Shengji Powder could accelerate chronic wound healing through the upregulation of VEGF.HIF-1α and Angl, which were closely related to angiopoiesis.
