兔牙移动中半胱氨酸蛋白酶抑制剂C表达与牙周骨改建的研究 被引量:1

摘要 目的:研究兔牙移动过程中半胱氨酸蛋白酶抑制剂C(cystatin C)在牙周组织中的表达变化,以及加力不同时间的变化特点。方法:选用20只2.0 kg体重的日本大耳兔建立正畸牙移动模型,加力0.8 N,分别在1、3、5、7、14 d后取龈沟液,ELISA检测龈沟液中cystatin C的含量,取龈沟液后,处死动物,HE染色观察牙周组织改建的变化,免疫组化SABC法检测cystatin C的表达随着时间变化的特点。结果:cystatin C在正畸牙移动初期(1~3 d)含量减少,破骨细胞增多,后期(5~14d)cystatin C含量增多,破骨细胞减少。结论:在正畸力的诱导下,cystatin C参与了正畸牙移动骨改建过程中有机基质的降解。 Objective: To investigate the expression of cystatin C during orthodontic tooth movement in rabbits.Methods: Experimental models of orthodontic tooth movement were established in 20 Japanese Big Ear Rabbits with about 2.0 kg weight.0.8 N orthodontic force was applied on the models.The gingival crevicular fluid was obtained and the content of cystatin C was measured by ELISA 1,3,5,7 and 14 days after the force application,then,the animals(n=4) were sacrificed respectively.The morphology was observed by HE staining.The cystatin C expression in the bone was observed and quantified with SABC.Results: In the early stage(at 1~3 d) of tooth movement cystatin C expression decreased and osteoclasts increased;in the late stage(at 5~14 d) the cystatin C expression increased and osteoclasts decreased.Conclusion: Cystatin C participates in proteolysis during orthodontic tooth movement.
