Correlation of Interleukin-10-1082G/A Single Nucleotide Polymorphism to the Susceptibility of Bladder Cancer

摘要 目的探讨白细胞介素-10(IL-10)基因启动子区域-1082G/A位点单核苷酸多态性与膀胱癌危险性的关系。方法选取浙江汉族人群中膀胱癌患者400例为病例组,400例健康体检者为对照组,以等位基因特异性扩增(allele specific PCRamplification,AS-PCR)结合DNA测序方法,检测病例组和对照组的IL-10-1082G/A基因型,比较不同基因型与膀胱癌危险性以及膀胱癌临床分期之间的关系。结果病例组和对照组的IL-10-1082G/A位点基因型分布和等位基因频率之间差异有统计学意义(χ2=8.375,P<0.05;χ2=8.322,P<0.01);AA基因型的个体与携带G等位基因型的个体(GG型和GA型)相比,携带等位基因A的个体与携带等位基因G的个体相比,其患膀胱癌危险性分别增加2.055倍和1.990倍(OR=2.055,95%CI:1.252~3.374;OR=1.990,95%CI:1.237~3.203);IL-10-1082G/A位点基因型与膀胱癌的临床分期无直接相关性。结论 IL-10-1082G/A多态性与膀胱癌危险性相关,该位点SNP检测可作为膀胱癌可能的分子标志物。 Objective To investigate the relationship between polymorphism at - 1082G/A in the promoter region of the IL - 10 gene and the risk of bladder cancer. Methods In a hospital based case - control study for the Chinese population in Zhejiang, a total of 400 patients with bladder cancer and 400 age - and sex - matched healthy controls subjects were collected. We analyzed the genotypes of IL - 10 - 1082G/A by allele specific PCR amplification ( AS - PCR) and DNA sequencing, and determined their association with bladder cancer risk and clinical stage. Results The genotype distribution and allele frequency of IL - 10 - 1082G/A gene had significant differ- ences between patient group and control group(x2 = 8. 375, P 〈 0.05 ;X2 = 8. 322, P 〈 0.01 ). Compared to the individual carrying AA genotype, the risk of bladder cancer with GG genotype and GA genotype increased 2. 055 times ( OR = 2. 055, 95% CI : 1. 252 - 3. 374). Compared to the individual carrying allele A, the risk of bladder cancer with allele G increased 1. 990 times( OR = 1. 990, 95% CI: 1. 237 -3. 203). We did not observe any association between the genotypes of IL -10 -1082G/A with the clinical stage of bladder canc- er. Conclusion IL - 10 - 1082G/A gene polymorphism may be a risk factor for bladder cancer and it may serve as a molecular marker for detecting bladder cancer.
