人骨髓间充质干细胞与血管内皮细胞的基因表达差异分析

摘要 目的探索人骨髓间充质干细胞向血管内皮细胞分化的分子生物学机制及调控靶标。方法应用基因表达谱芯片技术,检测人骨髓间充质干细胞和脐静脉内皮细胞的基因表达差异。结果在检测的3946条基因中,人骨髓间充质干细胞和血管内皮细胞非差异表达基因占84%。差异表达基因中,间充质干细胞高表达283条,低表达344条。这些基因可以划分为5个主要的功能群:①细胞骨架和运动蛋白基因;②细胞受体相关基因;③细胞信号和传递蛋白相关基因;④发育相关基因;⑤蛋白翻译、合成相关基因。结论人骨髓间充质干细胞有潜在的内皮分化能力,分化机制和分化调控与以上五类特异基因的表达有关。 Objective To study the molecular mechanisms and regulation targets of endothelial differentiation from human bone marrow mesenchymal stem ceils. Methods Genes differentially expressed between human bone marrow mesenchymal stem cells and human umbilical vein endothelial cells were analysed by using DNA microarray technique. Results Among 3946 genes screened, about 86% was of no difference. Among the differentially expressed genes, 283 were highly expressed in mesenchymal stem cells, and 344 were low expressed. These genes mainly had following functions : (1)cytoskeleton and motion protein - related genes ; (2)cell receptor - related genes ; (3)cell signal and transduction protein -related genes;(4)eell development- related genes;(5)protein translation and synthesis -related genes. Conclusion Human bone marrow mesenchymal stem cells is of great endothelial differentiation potential. The biological mecha- nisms and regulation targets of endothelial differentiation might be related to these specific genes expression.
