摘要
目的探讨联合偶合概率(RMP)与似然比率(LR)在炭化骨骼个人识别中的计算方法及意义。方法常压下以300℃对两例成人股骨焚烧1h后,液氮下研磨得到骨粉,乙二胺四乙酸(EDTA)脱钙,改良的酚-氯仿法提取DNA并纯化,5色荧光标记,PCR复合扩增,基因分析仪进行电泳,数据收集与分析。结果累积识别力(TDP,男):0.999 999 980 3,TDP(女):>0.999 999 999 999 99;RMP(男):2.322 5×10-12,RMP(女):4.931 3×10-24;LR(男):4.305 7×1011,LR(女):2.027 9×1023。结论 300℃炭化骨骼可提取到DNA,利用存留基因图谱比对及TDP、RMP、LR的计算可进行个人识别。
Objective To analyse random match probability ratio(RMP) and likelihood ratio(LR) in adult burning bone. Methods 2 adult femoral bone,female and male in half,were incinerated at 300℃ for 1 hour in normal pressure,then grinded to be bone meal in the liquid nitrogen before decalcification by ethylenediamine tetraacetic acid(EDTA). The deoxyribonucleic acid(DNA) was extracted by classical phenol and chloroform, subsequently the DNA was five-fluorescent labeted, amplificated by PCR,and the data was collected and analyzed by using genetic analyzer. Results The TDP value was 0. 999 999 980 3,and that of the female was 0. 999 999 999 999 99. The RMP value of the male was 2. 322 5×10-12 ,and that of the female was 4. 931 3×10-24. The LR of the male was 4. 305 7 × 1011 , and the female was 2. 027 9 × 1023. Conclusion The DNA can be extracted from the bone burned at 300℃ ,and the remaining gene locus atlas comparion,the TDP,RMP,LR value can be used for individual identification.
