缺血再灌注损伤大鼠脑组织细胞因子VEGF mRNA的变化及通心络对其的影响 被引量:1

The Expression of VEGF mRNA after Cerebral Ischemia-reperfusion Injury and the Effeet of Tongxinluo Capsule in Rats

摘要 目的探讨血管内皮生长因子(VEGF)mRNA在大鼠缺血/再灌注损伤(MCAO)脑组织的表达情况及通心络对其的影响。方法采用MCAO模型并予通心络灌胃,应用反转录集合酶链式反应(RT-PCR)检测缺血再灌注损伤后5d、7d、14d、21d、30d神经干细胞增殖分化相关细胞因子VEGF mRNA的变化。结果缺血再灌注模型组VEGF mRNA在不同时间段均有表达,VEGF mRNA在造模后第3天开始表达增强,第5天表达最高;给予通心络处理后第3天缺血侧VEGF mRNA开始表达增强,5d、7d、14d、21d、30d时PCR表达灰度值均高于缺血对照组。结论缺血/再灌注损伤大鼠脑组织细胞因子VEGF mRNA表达增强,通心络胶囊可强化此反应,进而可能诱导神经干细胞增殖。 Objective To explore the expression of vascular endothelial growth factor (VEGF) mRNA in cerebral ischemia- reperfusion injury in rat brain tissues and the effect of Tongxinluo capsule. Methods Sprague -Dawley(SD) rats after middle cerebral artery occlusion (MCAO) were perfused with Tongxinluo capsule. The neural stem cell proliferation and differentiation related cell factors of VEGF mRNA were detected after ischemiareperfusion injury at 5,7, t4,21 ,and 30 d by reverse transcriptase polymerase chain reaction (RT - PCR). Results VEGF mRNA of ischemia - reperfusion models was expressed in different period. The expression began to enhance at 3rd day,and it reached peak at fifth days. The express of ischemia side VEGF mRNA was enhanced after tongxinluo at 3d, and PCR express gray values were higher than that in the control group at 5,7,14,21, and 30 days. Conclusion The expression of could VEGF mRNA express was enhanced after ischemia - reperfusion injury in rat brain tissues. Tongxinluo capsule can strengthen the response and induce neural stem cell proliferation.
