生泰王对运动疲劳小鼠脑组织端粒酶表达的影响

摘要 目的观察生泰王对运动疲劳小鼠脑组织形态、脑组织的指数和端粒酶表达的影响,进而探讨生泰王增强体力、延缓衰老和干预运动疲劳损伤的作用机制。方法取昆明种小鼠随机分为4组,生泰王大、小剂量组分别灌胃给药生泰王饮剂5g/(kg.d),2.5g/(kg.d);对照组又分为运动组和非运动组,均灌胃等剂量的生理盐水,连续给药21d,于末次给药后1h除非运动对照组外,其他3组小鼠行游泳实验60min。处死小鼠取脑组织称重,并行HE染色检测脑组织形态学变化,免疫组化检测脑组织端粒酶的表达。结果生泰王可使力竭运动小鼠脑组织端粒酶表达下调,与力竭运动对照组相比有统计学意义(P<0.01);力竭运动对照组小鼠脑细胞形态多为三角形,锥体细胞数量显著减少;非运动对照组小鼠脑细胞形态圆润多为圆形或卵圆形,细胞间质比较紧密,血管壁光滑完整;生泰王大、小剂量组小鼠脑细胞形态圆形或卵圆形较多,而三角形较少,锥体细胞数量较多;脑组织的指数各组与力竭运动对照组相比有统计学意义(P<0.01)。结论生泰王通过下调力竭运动小鼠脑组织端粒酶表达,提高小鼠的抗运动疲劳能力,对力竭运动损伤具有干预的作用。 Objective To observe the effect of Shengtaiwang (STW) on cerebral morphology,brain tissue index,the expression Of telomerase in sports fatigue - mice. Methods Kunming mice were randomly divided into four groups: Large and small doses STW treatment group treated with STW,exercise group and non exercise group treated with isodose saline. After 1 hour of the last admin- istration,swimming test was performed for 60 rain. The brains were removed and weighted after the mice were killed. Morphology of the brain tissue was examined by HE staining. Immunohistochemistry was used to detect the expression of telomerase in brain. Results STW could decrease the expression of telomerase in brain tissue in exhaustive swimming mice compared with exercise control group (P〈0.01). Most of the brain cells were triangle- like and the amounts of pyramidale were obviously decreased in exercise control group. Most of the brain cells were round and elliptical, intercellular substance was dense, vascular wall was smooth and complete in non exercise control group. There were round and elliptical brain cells and less triangle - like brain cells in large and small doses groups. There were significant differences in the amounts of pyramidale compared with exercise control group(P〈0.01). Compared with the control group,index of brain tissue showed significant difference in every group(P〈0.01). Conclusion STW had a benefi- cial effect on sports fatigue injury by downregulating the expression of teiomerase in brain of exhausting mice and increasing the ability of anti sports fatigue.
