摘要
目的探讨少腹逐瘀丸对子宫内膜异位症大鼠异位子宫内膜组织中肿瘤坏死因子(TNF-α)和血管内皮生长因子(VEGF)mRNA表达的影响。方法采用自体移植法建立子宫内膜异位症大鼠模型,将造模成功的大鼠随机分为模型组、孕三烯酮组及少腹逐瘀丸高剂量组、低剂量组,并设假手术组。给药30 d后,以免疫组化法检测大鼠在位以及异位内膜组织中TNF-α的表达,以RT-PCR法检测VEGF mRNA表达。结果少腹逐瘀丸能降低子宫内膜异位症大鼠TNF-α的表达(P<0.05),显著降低VEGF mRNA的表达(P<0.01)。结论少腹逐瘀丸通过降低TNF-α和VEGFmRNA的表达,抑制子宫内膜细胞的异位黏附、种植和生长,这可能是其治疗子宫内膜异位症的作用机制之一。
Objective To investigate the influences of Shaofu Zhuyu Wan on expressions of endometrial tumor necrosis factor-α (TNF-α) and vascular endothelial growth factor (VEGF) mRNA in rats with endometriosis. Methods The rat model of endometriosis was established by using autotransplantation and then successfully modeled rats were randomly divided into model group, gestrinone group and high-dose and low-dose Shaofu Zhuyu Wan group (high-dose group and low-dose group ), and a sham-operation group was set in addition. After giving medicinals respectively for 30 days, the expression of TNF-α was detected in eutopie and ectopic endometrium by using immunohistomechemistry technique, and the expression of VEGF mRNA was detected by using RT-PCR. Results Shaofu Zhuyu Wan reduced the expression of TNF-α ( P 〈 0.05 ) and significantly reduced the expression of VEGF mRNA (P 〈0.01 ) in rats with endometriosis. Conclusion Shaofu Zhuyu Wan can inhibit ectopic adhering, implanting and growth of endometrial cells through reducing the expressions of TNF-α and VEGF mRNA, which may be one of mechanisms of Shaofu Zhuyu Wan in treatment of endometriosis.
