桔梗皂苷-D干预大鼠肺成纤维细胞纤维化的实验研究 被引量:2

摘要 目的 观察桔梗皂苷-D(PD)对转化生长因子(TGF-β1)诱导大鼠肺成纤维细胞纤维化的干预作用影响.方法 通过TGF-β诱导大鼠肺成纤维细胞纤维化,采用PD对细胞进行干预,检测大鼠肺纤维化细胞代谢活性,RT-PCR检测细胞信号蛋白Smad3、Smad7基因的表达,以及流式细胞术检测细胞上TGF-βII受体的表达.结果 干预组细胞代谢活性平均OD值0.299±0.022,模型组0.342±0.012(P<0.05);干预组Smad7表达较模型组上调,Smad3表达较模型组下降;干预组平均TGF-β II受体表达率(28.97±0.54)%,模型组(36.62±0.58)% (P<0.05).结论 PD可以抑制TGF-β诱导的大鼠肺成纤维细胞增殖和Smad3及TGF-β II受体表达,增强抑制性信号蛋白Smad7的表达. Objective To investigate the effect of Platycodin D (PD) in treating transforming growth factor(TGF- β ) mediated rat pulmonary fibrosis cells. Methods Metabolic activity of pulmonary fibrosis cell and the expression of signal protein Smad3,Smad7 and TGF-α II reception were detected. Results The mean A of metabolic activity was 0.299 ± 0.022 in intervention groups and 0.342 ± 0.012 in model group (P〈0.05). Compared with model group,intervention groups expressed lower Smad3 and higher Smad7 (P〈0.05). Pulmonary fibrosis cell expressed TGF- 13 II reception was 28.97 ± 0.54% in intervention groups and 36.62 ± 0.58% in model group (P〈0.05). Conclusion PD could obviously reducese pulmonary fibrosis by inhibited the expression of signal protein Smad3 and TGF- β II reception and promoted the expression of signal protein Smad7.
