摘要
目的研究叶酸水解制备蝶酸并建立测定蝶酸含量的方法。方法采用高效液相色谱法测定含量,色谱柱:PhenomexneC18(250 mm×4.6 mm),流动相为甲醇和水25︰75,水中含2%醋酸、0.2%三乙胺。流速1 ml/min。紫外检测器检测波长:285 nm处最大吸收。结果得到含量为73%的蝶酸粗品,蝶酸在1.6~9.6μg/ml有好的线性关系,相关系数为0.9991;稳定性实验结果 RSD=0.42%;样品加标回收率平均值为98.9%。结论建立的高效液相色谱法具有良好的精密度和准确度,适用于测定蝶酸的含量。
Objective: To determine pteroic acid in the samples synthesized in our laboratory by hydrolyzing folic acid. Methods: HPLC method was developed employing reserved-phase chromatographic column Phenomexne C18 (250 mm× 4.6 mm) and UV detection in the wave length of 285nm. The mobile phase consisted of methanol and water (containing 2% acetic acid, 0.2% triethylamine ) (25 : 75, v/v) with a flow rate of 1 ml/min. Results : The purity of synthesized pteroie acid was about 73%. The method provided a good linearity from 16 Ixg/mL to 9.6 p.g/ml. Relativity coefficient was 0. 9991, by the results of stability test RSD was 0.42%. The average recovery was 98.9%. Conclusion : The developed HPLC method is accurate and precise, which can be used for the content determination of pteroic acid.
