摘要
目的探讨脑胶质瘤中HINT1基因启动子区的甲基化状态。方法采用双酶切联合定量PCR法和MSP法检测70例脑胶质瘤和30例正常脑组织中HINT1基因启动子区域甲基化状况。结果经双酶切后实时荧光定量PCR和MSP法检测,正常脑组织和胶质瘤HINT1基因启动子区域呈非甲基化状态,与性别、年龄和病理分级无明显的相关性。结论脑胶质瘤中HINT1基因启动子区域呈非甲基化状态。
Objective To discuss the promoter methylation status of HINT1 gene in glioma. Methods We adopted the double digestion and the real-time quantitaty PCR method and MSP method to detect tlie methylation of HINT1 gene promoter of tissues. These tissues included glioma tissue of 70 patients and the normal brain tissue of 30 cases. Results The results of the double digestion and fluorescent quantitative real-time PCR and MSP Method demonstrated that HINTI gene promoter was unmethylated state, both of normal brain tissue and glioma tissue. There was no significant correlation between the unmethylated state of HINT1 gene promoter and gender, age, pathological types and pathologic stage. Conclusion HINT1 gene promoter of glioma is in the unmethylated state.
