摘要
从胎儿脑白质中提取 GFAP,分子量为54kd 及45kd。免疫兔得到兔抗人 GFAP抗血清,滴度为1∶8~1∶14,除与脑、背髓和星形细胞瘤有反应外,其它外周神经和组织器官均无反应。用间接免疫荧光和 PAP 染色,11种肿瘤细胞系中只有“BT_(325)”细胞为阳性。100例不同类型的脑肿瘤及其它部位肿瘤中只有星形细胞瘤、胶母细胞瘤和室管膜瘤出现阳性。
In this study glial fibrillary acidic protein(GFAP)was isolated from the white matter ofhuman fetal cerebrum by hydroxyapatite chromato-graphy.Specific antiserum against GFAP was ge-nerated by immunizing the rabbits with this orepara-tion.The antiserum reacted only with brain,spinalcord and astrocyttoma besides original GFAPpreparation.as demonstrated by immunodiffusion,immunoelectrophoresis,ELISA and immunoblotting,No cross reaction was observed with peripheralnerve and non-nervous tissues and organs.Eleventumor cell lines and sections of 100 cases oftumors from brain and other tissues were stainedwith the antiserum by immunofluorescence andPAP method.Among the cell lines only BT325,a human glioblastoma cell line,gave positivereaction,Among the tumors studied only astrocytomas,glioblastomas and ependymomaswere positive,and other tumors were negative.All the results demonstrated that this antiserumis specific against GFAP,and can be used inbiological studies of CNS and in differentialdiagnosis of brain tumors.
