摘要
应用环境扫描电子显微镜和能谱分析方法研究转广谱抗真菌基因碱性几丁质酶基因RC24、苜蓿葡聚糖酶基因β-1,3-Glu、酸性几丁质酶基因RAC22对不同生长期非转基因水稻(七丝软占)和转抗真菌水稻(双价抗真菌转基因水稻转品8、三价抗真菌转基因水稻转品1)抗病元素化学元素组成差异。分析结果表明:除C、H、O外,水稻叶、叶鞘主要含有Si、K、Na、Cl、Mg5种与抗病相关的化学元素,其中Si的相对含量为10%~15%,K、Na相对含量为0.2%~3.6%,Mg、Cl相对含量较低,大部分样品小于1%。外源基因的转入对水稻叶、叶鞘化学元素相对含量有一定影响,其中Si、K、Na的相对含量变化明显,差异最明显的分别达50.8%,115.3%,251.9%。存在时空变化,其机理尚待研究。
Relative content of elements in leaf and leaf sheath of Oryza sativa L.,including Qisiruanzhan(non-transgenic variety),Zhuanpin 1(trivalent transgenic variety) and Zhuanpin 8(double-transgenic variety) were analyzed with ESEM and X-ray EDS.Results showed that besides C,H,O,elements for disease resistance contented in Oryza sativa L.were as followed: Si, K,Na,Cl,Mg.Content of Si is 10%~15%,while is 0.2%~3.6% for K and Na.Content of Mg and Cl are lowest,which is below 1%.Expression of foreign gene would affect content of elements of Oryza sativa L.Content of some elements between transgenic Oryza sativa L.and non-transgenic ones are different,among which the content of Si,K,Na vary obviously.And the difference among them is of a space and time character.
