星形细胞瘤细胞DNA定量研究的临床生物学意义

Clinical and biological significance of quantitative study of cellular DNA content in astrocytoma

摘要 本研究应用图像分析技术对54例星形细胞瘤细胞 DNA 含量进行了定量分析。结果显示,瘤细胞 DNA 含量明显增高,瘤细胞 DNA 含量及干系水平与病理分级、临床预后、肿瘤大小及囊性变均有显著的正相关性,DNA 含量越高,则恶性程度越高,预后越差。细胞 DNA 的定量分析较病理分级更为客观准确地反映了星形细胞瘤的生物学特征,为其诊断、治疗、恶性程度判断及预后估价提供重要的信息。 In the present study,celluar DNA contentwas quantitatively determined in 54 specimens fromastrocytoma by means of image analysis technology.Results showed that cellular DNA content ofastrocytoma was obviously higher than that ofnormal brain tissue.It can be classified as D/ND,LAN,HAN astrocytoma according to the level ofits cellular DNA stem line.Cellular DNA contentand DNA stem line of astrocytoma were positivelycorrelated with its gradation of malignancy,clinicalprognosis,tumour size and cyst-formation.CellularDNA content and DNA stem line reflected thebiological features of astrocytoma more objectivelyand accurately.The higher the cellular DNA con-tent was,the higher the tumour's malignant degreewas,and the worse the prognosis was.It wasobserved the prognosis of D/ND,LAN astrocytomawas much better than that of HAN astrocytomaeven in the same pathological grade.This papersuggest that the quantitative study of cellular DNAcontent of astrocytoma can provide valuable comple-ment to conventional pathological diagnosis andjudge the clinical prognosis more exactly.It serveas an objective and relative independent targetwill be of great importance in the clinical andbasical study of astrocytoma.
