摘要
利用本室建立的 NHG-1第55~60代荷人脑胶质瘤裸小鼠共190只进行一种新的抗恶性胶质瘤药物筛选的方法学研究,采用荷瘤鼠腹腔一次注射~3H-TdR30μCi,18小时后取肿瘤、小肠和股骨制成液闪均相液,测氚的量。此法可客观反映被测组织中细胞 DNA 的合成能,在抗癌药作用下,既能观察肿瘤细胞增殖受抑的程度,又能反映荷瘤鼠肠上皮和骨髓细胞的受损情况。实验周期,从治疗开始仅需4天。
A new screening method for anti-glioma agentswas developed.30μCi^3 H-thymidine was injected tothe abdominal cavity of bearing human malignantglioma xenografts nude mice(NHG-1 model).18hours later the metastatic tumor,in femur boneand small intestine were removed and then the CPMwas measured in a homogeneous scintillation coun-ting.This is a rapid and accurate method forassaying of DNA synthesis of cells,it may beuseful in estimating tumor response to chemothe-rapy as well as side effects of anti-cancer agentson the bone marrow and small intestines.4 anti--cancer agents were studied in this method onmalignant glioma.The end sesults was similar tothat of conventional methods.
