摘要
目的建立高效液相色谱法测定葛花中葛花苷含量方法。方法采用依利特Hypersil ODS(4.6 mm×150mm,5μm)色谱柱;流动相为乙腈-0.1%磷酸(17∶83),流速为1mL.min-1,柱温为35℃,检测波长:265nm,进样量:10μL。结果葛花苷在0.102 4~1.229mg.mL-1内与峰面积线性关系良好,平均回收率为100.9%,RSD为1.7%。结论所建立之方法准确可靠、灵敏度高、专属性强,可用于测定葛花中葛花苷的含量。
Objective To establish an HPLC method for the determination of kakkalide in Pueraria lobata Ohwi. Meth- ods Hypersil ODS (4. 6 min×150 mm, 5 μm) was used with acetonitrile-1% phosphoric acid solution as the mobile phase. The flow rate was 1.0 mL·min 1 and the column temperature was 35℃. The detection wavelength was 265 nm and the injection volume was 10μL. Results The linear range of kakkalide was 0. 102±1. 229 mg·mL-1. The average recovery was 100.9%, with RSD was 1.7%. Conclusion The method is reliable, accurate and specific with good reproductivity and can be used for the content determination of kakkalide in Pueraria lobate Ohwi.
