摘要
目的:探讨丹酚酸B对糖尿病大鼠肺组织中肾素-血管紧张素系统活性的影响。方法:取4周龄健康SD大鼠40只,分为正常对照组、模型组、厄贝沙坦组、丹酚酸B组。分别给予胃管灌注生理盐水(正常对照组、模型组)、厄贝沙坦15 mg/kg(厄贝沙坦组)、丹酚酸B 5 mg/kg(丹酚酸B组),1次/天,每周灌胃5次。8周后处死大鼠,留取肺组织进行组织匀浆,测定血管紧张素II(Angiotensin II,AngII)浓度,通过HE染色观察肺组织病理变化,免疫组化和图像分析法检测肺组织中血管紧张素II的I型受体(Angiotensin II receptor I,ATIR)的表达。结果:1.糖尿病模型组肺组织肺泡萎陷、破坏,肺泡间隔增宽。厄贝沙坦组、丹酚酸B组大鼠肺泡结构改善。2.血管紧张素II浓度:模型组、厄贝沙坦组、丹酚酸B组均显著高于对照组(P<0.05),厄贝沙坦组与模型组相比无差异(P>0.05)、丹酚酸B组低于模型组(P<0.05)。3.ATIR的表达:模型组高于对照组(P<0.05),厄贝沙坦组、丹酚酸B组低于模型组(P<0.05)。结论:糖尿病大鼠肺组织中肾素血管紧张素系统活性增加,可能参与了肺组织的损害;丹酚酸B、厄贝沙坦能降低ATIR表达,改善肺结构。
Objective: To investigate the effect of Salvianolie acid B on Renin-angiotensin system activity in diabetic rat lung. Methods: Heath SD rats were divided into normal group, model group, Irbesartan group , Salvianolic acid B group, and were admin- istered saline (normal and model group), Irbesartan 15mg/kg, Salvianolic acid B 5mg/kg, once per day, five times per week. Rats were killed after 8 weeks, lung tissue was homogenated, Angiotensin II was detected. Pathological changes in lung tissue was observed through HE staining. Expression of Angiotensin II receptor I (ATIR) was measured by Immunohistochemistry. Results: 1. Alveolars were collapsed, destructed, alveolar septum was widened in diabetic model rats. Alveolar structure was improved in Salvianolie acid B and Irbesartan group. 2. Angiotensin II concentration: Angiotensin II concentrations in diabetic model , Salvianolic acid B and Irbesar- tan group were higher than that in normal group ( P 〈 0. 05 ) . No differences were noted in Irbesartan and diabetic model group ( P 〉 0. 05), Angiotensin II levels in Salvianolic acid B group were higher than that in model group ( P 〈 0. 05 ) . 3. Expression of ATIR : The expression of ATIR in diabetic model group was higher than in normal rats ( P 〈 0. 05 ), in Salvianolie acid B and Irbesartan group was less than that in diabetic model group ( P 〈 O. 05) . Conclusion: Renin-angiotensin system activity elevated in diabetic rat lung tissue, which may participate in the lung damage. Salvianolie acid B and Irbesartan reduced it and improved lung structure.
