丹参为主的中药舒肝颗粒对活化大鼠肝星状细胞表达转化生长因子β_1的影响

Effect of Shu-gan Granule on Expression of TGF-β_1 of Hepatic Stellate Cells in Rat

目的:观察舒肝颗粒对活化大鼠肝星状细胞表达转化生长因子TGF-β1的影响,以探讨舒肝颗粒对大鼠肝星状细胞活化的影响及相关机制,为舒肝颗粒临床应用提供进一步理论依据。方法:用空白对照组,0.01、0.02、0.04 mg/mL的舒肝颗粒分组处理活化的HSC 24 h后,用放射免疫法测定各组HSCTNF-α表达情况,荧光定量PCR法测定各组HSC TGF-β1表达情况。结果:用荧光定量PCR法测各组TGF-β1的表达情况:空白对照组、舒肝颗粒0.01 mg/mL组、舒肝颗粒0.02 mg/mL组及舒肝颗粒0.04mg/mL组TGF-β1表达水平通过计算2-△△Ct值分别为:(1.00±0.09)、(0.71±0.08)、(0.55±0.07)、(0.44±0.03),且组间差异有统计学意义(P<0.01)。结论:舒肝颗粒可通过抑制HSC TGF-β1的表达,发挥其抗纤维化作用。

Objective： To research the affect of shu-gan granule on expression of TGF-β1 on activation of Hepatic Stellate Ceils in Rat activated invitro, investigate the main mechanism of shu-gan granule in protection of liver fibrosis. Methods： The experiment was divided into the blank control group, shu-gan granule 0. 01 mg/mL group, shu-gan granule 0. 02 mg/mL group and shugan gran- ule 0. 04 mg/mL group. HSC was intervented in vitro for 24h, then detected TGF-β1 expression by fluorescence quantitative PCR. Re- suits： The numerical of 2-△△Ctof control group and each drug intervention group were： （1.00 ± 0. 09）, （0. 71 ± 0. 08）, （0.55 ±0. 07）, （0. 44 ±0. 03）, and there were statistical significance among them （P 〈0. 01 ） . Conclusion： Shu-gan granule can resist hepatic fibrosis through inhibiting the expression of HSC TGF-β1.
