微波制备IFN-γ处理的小鼠H22肝癌瘤苗免疫荷瘤鼠的抗瘤作用

摘要 目的探讨用微波方法制备干扰素-γ(interferon-γ,IFN-γ)处理的小鼠H22肝癌细胞瘤苗(microwave and IFN-y treated H22 tumor cells making H22 tumor vaccine,MITTV-H22)激发机体的抗肿瘤免疫反应。方法小鼠皮下接种H22肿瘤细胞,同时于肿瘤接种部位皮下接种MITTV-H22进行免疫治疗。观察2组小鼠的生存期,治疗结束后取瘤称质量、测瘤体积、计算体积抑瘤率和质量抑瘤率、观察瘤组织病理切片。结果①实验组小鼠的平均生存期为46.5d,对照组小鼠的平均生存期为34.0d(P<0.05)。②实验组小鼠的瘤块体积增长速度明显慢于对照组(F=179.318,P<0.0)。③实验组小鼠的平均瘤体积、平均瘤质量小于对照组。④实验组的体积抑瘤率为(57.80±18.40)%,质量抑瘤率为(62.65±23.53)%。⑤实验组肿瘤组织坏死明显增多,大片坏死组织中可见散在成团或岛屿状的残留瘤细胞,淋巴细胞多见,瘤体周边纤维母细胞明显增生。对照组肿瘤组织中瘤细胞增生活跃,局部可见小片状坏死灶,肿瘤细胞间淋巴细胞极少见,瘤体周边有很少或未见纤维母细胞增生。结论 MITTV-H22对H22细胞型肝癌具有较强的治疗作用。
