摘要
目的 观察大鼠坐骨神经挤压伤后纤溶成分中组织型纤溶酶原激活剂 (t PA)和纤溶酶原激活剂抑制因子 1(PAI 1)蛋白表达的变化 ,了解t PA、PAI 1在周围神经损伤后修复过程中的作用。方法 取 2 1只Wistar大鼠 ,分别造成坐骨神经挤压伤模型。在术后 6h、1d、3d、7d、14d和 2 1d分别取材进行组织学、免疫组化观察。结果 术后 3d时神经断端回缩球处的t PA抗原呈阳性反应。 7d时损伤处的雪旺细胞明显增多 ,且t PA、PAI 1均有表达。 14d、2 1d时t PA表达有所减少 ,而PAI 1的表达则逐渐增多。结论 t PA和PAI 1参与周围神经挤压伤后的病理生理改变 ,在损伤神经修复过程中具有一定的作用。
Objective To observe the changes of tissue type plasminogen activator(t PA) and plasminogen activator inhibitor 1 ( PAI 1) expression in adult rat sciatic nerve after crush injury, and investigate the effects of t PA and PAI 1 on peripheral nerve injury and regeneration. Methods Sciatic nerve crush injury model was set up in 21 Wister rats. Histological and immunohistochemical examination of the nerve samples were performed 6 hours, 1 day, 3 days, 7 days, 14 days, and 21 days after the operation. Results The t PA antibody was positive in the retraction bulb of the proximal segments of crushed sciatic nerve after 3 days. There was enormous proliferation of Schwann cells in the lesion area. Both t PA and PAI 1 were expressed 7 days after the injury. From then on, the expression of t PA decreased whereas the expression of PAI 1 increased gradually. Conclusions T PA and PAI 1 not only participate in the pathophysiological changes of peripheral nerves after crush injury, but have some effects on peripheral nerve repair as well.
