摘要
目的探讨血管生成与血小板源生长因子的表达在大肠癌的发生、发展中的作用.方法采用免疫组化 SABC 法,对45例大肠癌患者和10例正常大肠组织标本进行微血管密度和 PDGF 表达的检测,微血管的染色结果用图象分析系统进行处理,并结合大肠癌的临床病理进行分析.结果 PDGF 染色阳性29例,占64.4%,其中强阳性10例,占22.2%,而在正常的大肠组织中没有1例 PDGF 的表达阳性.血管生成与大肠癌的局部浸润程度(0.32±0.09 vs 0.29±0.09)、Dukes 分期(0.36±0.06 vs 0.25±0.05)、器官转移情况相关(0.38±0.06 vs 0.27±0.08).PDGF 表达阳性组的血管生成较 PDGF 表达阴性组明显增高(0.31±0.08 vs 0.25±0.06).结论 PDGF 是肿瘤血管生成的正性调节因子,血管生成在大肠癌的发展、演进中起重要的作用.
AIM To study the angiogenesis and PDGF expression in the colorectal carcinoma and to evaluate its clinical and pathological significance. METHODS Specimens of 45 colorectal carcinomas and 10 normal intestinal tissues were examined for PDGF expression and microvessel density by immunohistochemical staining,using antibody to PDGF and anti-Ⅷ factor related antigen antibody,the MVD staining results were analyzed by computer-aided image analysis system. RESULTS In the colorectal carcinoma patients,the positive rate of PDGF expression was 64.4% (29/45),and there was no PDGF expression in the normal intestinal tissues.There was a significant correlation between the angiogenesis and some clinico-pathological factors such as tumor infiltration (0.32±0.09 vs 0.29±0.09),organ metastasis (0.38±0.06 vs 0.27±0.08) and Dukes stage (0.36±0.06 vs 0.25±0.05).The MVD in the PDGF positive-expression group was higher than the PDGF negative-expression group (0.31±0.08 vs 0.25±0.06). CONCLUSION PDGF is an angiogenis factor and angiogenesis plays a very important role in the progess of human colorectal carcinoma.
