高效液相色谱法测定春根藤中的齐墩果酸和熊果酸 被引量:2

Determination of Oleanolic Acid and Ursolic Acid in Alyxia sinensis by HPLC

摘要 采用超声辅助提取,建立春根藤中齐墩果酸和熊果酸的高效液相色谱的分析方法。采用Kromasil C_(18)柱(4.6mm×250mm,5μm)色谱柱,以甲醇:水:磷酸(90:10:0.1,V/V/V)为流动相,流速1.0mL·min^(-1),检测波长210nm,柱温30℃。齐墩果酸和熊果酸进样量分别在0.484—4.84μg(r=0.9999)和0.664—6.64μg(r=0.9998)范围内与峰面积呈良好的线性关系,平均加标回收率(n=6)分别为97.5%和96.9%,RSD分别为1.5%和1.4%。该方法简便、准确、可靠,可用于春根藤中齐墩果酸和熊果酸含量的测定。 The method for the determination of oleanolic acid and ursolic acid in Alyxia sinensis by high performance liquid chromatography(HPLC) with extraction by ultrasonic-assisted extraction was established.The compounds were separated by Kromasil C18 column(4.6 mm×250 mm,5μm) with methanol-water-phosphoric acid(90:10:0.1,V/V/V) as mobile phase at a flow rate of 1.0mL·min-1,and the detection wavelength was 210nm,and the column temperature was maintained at 30℃.There were good linear relationships between the sample sizes of oleanolic acid and ursolic acid and peak areas in the ranges of 0.484-4.84μg(r=0.9999) and 0.664-6.64μg (r=0.9998) 〉 respectively,and the average additional standard recoveries of both were respectively 97.5%and 96.9%(n=6),and RSDs were 1.5%and 1.4%respectively.The method is accurate, reliable,and can be applied to determining the contents of oleanolic acid and ursolic acid in Alyxia sinensis.
