软肝宁抗四氯化碳诱导的大鼠肝纤维化 被引量:8

Experimental Study of Ruanganning in Rats with Counteracting Hepatic Fibrosis Induced by Carbon Tetrachloride

摘要 目的:探讨软肝宁对四氯化碳(CCl4)诱导的实验性肝纤维化大鼠纤维化水平的影响。方法:将60只大鼠随机分为6组:正常对照组,给予生理盐水。模型组,给予20%四氯化碳花生油溶液按10 mL.kg-1sc,首剂加倍,注射1次/5 d,首次注射后,用生理盐水按10 mL.kg-1ig,1次/d,连续6周。秋水仙碱组(ig 0.1 mg.kg-1)。软肝宁高、中、低剂量组(ig 0.12,0.06,0.03 g.kg-1)按损伤模型组同法给予CCl4造模,然后给予软肝宁给药,1次/d,连续给药6周后检测大鼠血清中的谷丙转氨酶(ALT)、谷草转氨酶(AST)、白蛋白(ALB)、血清透明质酸(HA)、层黏连蛋白(LN)、Ⅲ型前胶原(PCⅢ)和羟辅氨酸(HYP),并观察大鼠肝组织常规病理改变。结果:软肝宁能改善肝纤维化大鼠的肝功能,具有剂量依赖性地使升高的血清ALT,AST水平降低(P<0.05或P<0.01),使降低的ALB水平明显升高(P<0.05或P<0.01);软肝宁高、中剂量可显著降低血清HA,LN,PCⅢ水平和肝组织中过高的HYP(P<0.05或P<0.01);同时常规病理也提示,软肝宁中、高剂量组可明显减轻CCl4所致的大鼠肝细胞变性、坏死及肝组织损害程度。结论:软肝宁对CCl4诱导的大鼠肝纤维化有良好的防治作用。 Objective:To investigate the effects of Ruanganning on fibrosis level in in rats with liver fibrosis.Method:Sixty rats were randomly divided into six groups: the normal group,the model group,bifendate group(0.01 g·L-1),high Ruanganning group(0.12 g·kg-1),middle Ruanganning group(0.06 g·kg-1) and low Ruanganning group(0.03 g·kg-1).The treatment groups were orally administered once per day for six weeks,whereas the normal and model groups were orally administered with saline.Six weeks later,the blood simple was collected for the examination of alanine aminotransferase(ALT),aspartate aminotransferase(AST),albumin(ALB),hyaluronic acid(HA),Laminin(LN),precollagen typeⅢ(PCⅢ),hydroxyproline(HYP) and liver specimen were collected for the observation of pathology.Result: Compared with model group,Ruanganning could protect hepatic function.Following the treatment with Ruanganning,ALT and AST activities were significantly reduced and ALB activity was obviously increased in treating group(P0.05,P0.01),especially the middle and high doses could significantly degrade the serum levels of HA,LN,PCIII and the exorbitant HYP in liver tissue(P0.05,P0.01).Furthermore,pathological examination indicates Ruanganning can protect rats from hepatic fibrosis.Conclusion: Ruanganning can greatly protect from liver fibrosis in rats induced by CCl4.
