补肾法、疏肝法对促性腺激素预处理小鼠排卵过程组织蛋白酶-L蛋白影响的比较研究

摘要 目的:探讨补肾法、疏肝法对小鼠排卵关键蛋白水解酶Cat L蛋白的影响。方法:选择体重16-18g未成年小鼠随机分为正常组、对照组、疏肝组和补肾组超促排卵,采用Western blot、免疫组化方法检测注射hCG后0h、4h、8h、12h Cat L蛋白在各组小鼠卵巢的表达强度。结果:注射hCG后4h与对照组比较,疏肝组、补肾组小鼠卵巢Cat L蛋白表达均升高(P<0.01);注射hCG后8h与对照组比较,疏肝组小鼠卵巢Cat L蛋白表达升高(P<0.01),补肾组无统计学差异(P>0.05);注射hCG后12h与对照组比较,补肾组小鼠卵巢Cat L蛋白表达升高(P<0.01),疏肝组无统计学差异(P>0.05)。结论:补肾法通过使卵巢Cat L蛋白表达增强,使卵泡壁破裂而达到促排卵的作用,疏肝法通过使卵巢CatL蛋白表达迅速达高峰起到促进排卵的目的。 Objective:To detect the effect of kidney-tonifying treatment and liver-soothing treatment on cathepsin-L,a representative proteinase during ovulation.Methods:Immature 16~18g mice were primed with normal group,control group,liver-soothing treatment group and kidney-tonifying treatment group,and the expression of cathepsin-L protein was detected by Western blot and immunohistochemical at 4,8 and 12h after induction of ovulation by injecting the primed animals with 5 IU hCG.Results:Compared with control group,the expression of cathepsin-L protein was increased in liver-soothing treatment group and at 4h after injecting hCG(P〈0.01),and was increased in liver-soothing treatment group at 12h after injecting hCG(P〈0.01),but no statistically significant differences in kidney-tonifying treatment group at 12h after injecting hCG(P〈0.05),and was increased in kidney-tonifying treatment group at 12h after injecting hCG(P〈0.05),but no statistically significant differences in liver-soothing treatment group at 12h after injecting hCG(P〈0.05).Conclusion:Kidney-tonifying treatment promoted ovulation by enhancing the expression of cathepsin-L protein,and that of liver-soothing treatment by advancing the expression of cathepsin-L gene.
