摘要
目的:探讨黄芪煎剂治疗糖尿病肾病的机制。方法:应用链脲佐菌素建立大鼠糖尿病肾病模型,成模大鼠随机分为模型组、黄芪煎剂组,另设正常对照组。12周干预期后,分别检测各组:①肾皮质匀浆SOD、GSH-PX、MDA含量。②肾小球足突平均宽度、足突融合率、基底膜平均厚度。结果:与正常组相比较,模型组和黄芪煎剂组大鼠肾皮质GSH-PX、SOD降低、MDA升高(P<0.05),足突宽度、足突融合率、基底膜平均厚度升高(P<0.05);黄芪组与模型组相比较,上述指标改善(P<0.05)。结论:黄芪煎剂可以改善DN大鼠氧化应激,降低足突宽度、足突融合率、肾小球基底膜平均厚度。黄芪煎剂可能通过改善氧化应激状态,从而实现对肾小球超微结构的保护作用。
Objective:This paper aims to study the mechanism of action of Huangqi decoction in treating diabetic nephropathy(DN).Methods:Rat models with DN were prepared with Streptozotocin,which were then randomly divided into model group and Huangqi group.Another normal control group was also prepared.The following indicators were measured for each group after a 12-week intervention therapy:(1) the content of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and malondialdehyde(MDA) in the renal cortex;(2) the mean width of the podocyte foot processes(FPW),foot process fusion and average thickness of glomerular basement membrane(GBM).Results:A decrease in GSH-PX and SOD and an increase in MDA in the renal cortex(P〈0.05) as well as an increase in FPW,foot process fusion and average thickness of GBM(P〈0.05) were observed in both the model group and Huangqi group compared with those in the normal control group.But the above indicators improved after Huangqi decoction intervention.Conclusion:Huangqi decoction can improve the oxidative stress(OS) and reduce FPW,foot process fusion and average thickness of GBM in the DN rats.Therefore,Huangqi decoction might protect the glomerular ultrastructure through improving its oxidative stress.
