摘要
目的:通过观察观察前列煎对实验大鼠前列腺组织IL-1β、COX-2表达的影响,探讨前列煎治疗慢性非细菌性前列腺炎的可能机制。方法:参照Seethalakshm等的方法建立慢性非细菌性前列腺炎大鼠模型,将3月龄雄性Wistar大鼠随机分为正常组、模型组、治疗组、对照组,每组10只。治疗组予以前列煎水煎剂(含生药1.13g/mL),对照组予以前列通瘀水溶液(含生药0.06g/mL),均按1mL/100g体重灌胃;正常组和模型组予以等量的生理盐水灌胃。将各组大鼠干预30天,第30天称重后脱颈处死,取出前列腺并称湿重,免疫组化法检测前列腺组织IL-1β、COX-2表达。结果:模型组大鼠前列腺组织IL-1β、COX-2表达水平升高,与正常组比较,差异均有统计学意义(P<0.01)。治疗组和对照组大鼠前列腺组织IL-1β、COX-2表达水平较模型组均明显降低(P<0.01或P<0.05)。治疗组IL-1β、COX-2表达水平均低于对照组(P<0.05)。结论:前列煎可下调与炎症反应和组织纤维化密切相关的IL-1β、COX-2表达,这可能是前列煎治疗慢性非细菌性前列腺炎的机制之一。
Objective:To explore the possible mechanism of Qianliejian by the expressions of IL-1β and COX-2 protein in experimental chronic abacterial prostatitis rats prostate tissue.Methods:The experimental chronic abacterial prostatitis rat models were induced refered to Seethalakshm's method.And 3-month-old male Wistar rats were divided into four groups randomly:the normal group,the model group,the treat group and the control group,with 10 cases in each group.The treat group was given Qianliejian decoction 1 milliliter per 100 gram avoirdupois(comprise crude drug 1.13 gram per milliliter);the control group was given Qianlietongyu decoction 1 milliliter per 100 gram avoirdupois;the normal group and the control group were given the same dosage of normal saline.All animals were killed after 30 days' treatment,and prostate glands were obtained and the expressions of IL-1β and COX-2 protein were measured with the method of immunohistochemistry.Results:The expressions of IL-1β and COX-2 protein were markedly increased in the model group,compared to the normal group,the difference had statistical significance(P〈0.01).Compared to the model group,the expressions of IL-1β and COX-2 protein in treatment group and control group were markedly decreased,the difference had statistical significance(P〈0.01,P〈0.05).The expressions of IL-1β and COX-2 protein in the treatment group were fewer than that in the control group(P〈0.05).Conclusion:Qianliejian can decrease the expressions of IL-1β and COX-2 which are closely related to inflammation and fibroplasias,this may be one of the mechanisms of Qianliejian treating chronic abacterial prostatitis.
