摘要
目的:探讨伤科接骨片在下颌骨缺损修复中的作用机制。方法:健康雄性新西兰兔72只,随机分为正常对照组(24只)、模型对照组(24只)和伤科接骨片组(24只),建立下颌骨缺损模型。正常对照组、模型对照组动物给予正常饲料,伤科接骨片组动物给予伤科接骨片饲料。分别于建模后7、14、28、56天处死相同例数动物(n=6),收集下颌骨缺损处骨组织块。采用免疫组织化学技术检测骨保护素(Osteoprotegerin,OPG)、骨保护素配体(Osteoprotegerin ligand,OPGL)表达变化。结果:与正常对照组比较,模型对照组OPG、OPGL表达分别于建模后第7、14天显著提高(P<0.05),伤科接骨片组OPG表达于建模后第7、14、28、56天均显著提高(P<0.05),OPGL表达则提高不明显(P>0.05);与模型对照组比较,伤科接骨片组OPG表达于建模后第14、28、56天均显著提高(P<0.05),OPGL表达于建模后第14天显著降低(P<0.05),OPG/OPGL比值在建模后第14、28、56天显著上升(P<0.05)。结论:伤科接骨片可能通过影响OPG、OPGL的表达而促进下颌骨缺损修复。
Objective:To explore the repair mechanism of Shangke Jiegu tablets in mandibular defect.Methods:72 healthy male New Zealand rabbits were randomly divided into the normal control group(24),the model control group(24)and the Shangke Jiegu tablets group(24),then the mandibular defect model was established.Animals in the normal control group and the model control group received normal forage,animals in the Shangke Jiegu tablets group received Shangke Jiegu tablets forage.On the days 7,14,28 and 56 after establishing model,6 rabbits were killed separately.At the same time,the bone in the zone of mandibular defect was collected.The expressions of Osteoprotegerin(OPG)and Osteoprotegerin ligand(OPGL)were investigated by means of immunohistochemical technique.Results:Compared with the normal control group,the degree of expressions of OPG and OPGL increased remarkably on the days 7 and 14 after establishing model in the model control group separately(P〈0.05),the degree of expression of OPG increased remarkably on the days 7,14,28 and 56 after establishing model(P〈0.05)and the degree of expression of OPGL did not(P〈0.05)in the Shangke Jiegu tablets group;compared with the model control group,the degree of expression of OPG increased remarkably on the days 14,28 and 56 after establishing model(P〉0.05)and the degree of expression of OPGL decreased remarkably on the day 14 after establishing model(P〈0.05)in the Shangke Jiegu tablets group,the ratio of OPG/OPGL increased remarkably on the days 14,28 and 56 after establishing model(P〈0.05).Conclusion:Shangke Jiegu tablets may influence the expressions of OPG and OPGL so as to promote mandibular defect repairing.
