清肺饮对呼吸道合胞病毒感染大鼠血清白细胞介素-4、干扰素-γ及肺组织的影响

Effects of Qinfei Decoction on Serum Levels of IL-4 and INF-γ and Pathological Changes in Lung Tissue of Rats Infected by Respiratory Syncytial Virus

目的:观察清肺饮对呼吸道合胞病毒感染大鼠血清白细胞介素-4、干扰素-γ及肺组织影响。方法:42只SD大鼠被随机分成正常对照组、病毒感染模型组、中药高剂量组、中药中剂量组、中药低剂量组、和地塞米松对照组6组,大鼠在戊巴比妥钠腹腔注射麻醉下用呼吸道合胞病毒滴鼻吸入感染的方法建造病毒感染模型。采用荧光免疫试剂检测法,检测白细胞介素-4、干扰素-γ。观察肺组织病理学变化。结果:与正常对照组相比,病毒感染模型组大鼠白细胞介素-4升高、干扰素-γ下降,均有统计学差异(P<0.05,P<0.05),与病毒感染模型组比较,中药高、中、低剂量组及地塞米松组白细胞介素-4含量均明显减低(均P<0.01),中药中、低剂量组干扰素-γ含量升高(P<0.05,P<0.01),地塞米松组干扰素-γ含量升高(P<0.05)。清肺饮能减轻呼吸道合胞病毒感染大鼠肺泡壁增厚程度,减少炎症细胞浸润。结论清肺饮对呼吸道合胞病毒感染大鼠的血清白细胞介素-4有抑制作用、干扰素-γ有升高作用。清肺饮能减轻呼吸道合胞病毒感染大鼠肺组织损害程度。

Objective：To observe the effects of Qinfei Decoction on serum levels of IL-4 and INF-γ and pathological changes in lung tissue of rats infected by respiratory syncytial virus.Methods：42 rats were randomly divided evenly into normal control group,RSV infection model group,high dose Qinfei Decoction group,middle dose Qinfei Decoction group,low dose Qinfei Decoction group,and DXM group.To establish the model,rats were anesthetized by intraperitoneal injection in pentobarbite.Rats were dropped in nasal cavity with RSV,the contents of IL-4 and INF-γ were detected with enzyme linked immunosorbent assay（ELISA）.The pathological changes in lung tissue were observed.Results：Compared with the normal control group,the contents of IL-4 in the RSV infection model group were elevated,the contents of INF-γ in the RSV infection model group were lowered,the differences had significance（P〈0.05,P〈0.05）.Compared with the RSV infection model group,the contents of IL-4 in the high,millde and low dose Qinfei Decoction group and DXM group were lowered significantly（all P〈0.01）,the contents of INF-γ in the middle and low dose Qinfei Decoction group were elevated（P〈0.05,P〈0.01）,and that in DXM group was elevated（P〈0.05）,Qinfei Decoction could reduce the rat＇s alveolar wall thickening and inflammatory cell infiltration infected by RSV.Conclusion：Qinfei Decoction can reduce the serum content of IL-4,elevate the content of INF-γ in rats infected by RSV.Qinfei Decoction can reduce the degree of lung histologic lesion in rats infected by RSV.
