摘要
目的研究氧化苦参碱对人体骨肉瘤细胞(MG-63)凋亡的影响。方法在培养液体中加入不同浓度的氧化苦参碱(Oxymatrine,OM)进行不同时间长度的诱导,以流式细胞术法检测人体骨肉瘤细胞(MG-63)增殖、周期及凋亡情况。结果1~50μmol/L氧化苦参碱均能抑制骨肉瘤细胞增殖,同时能够阻滞MG-63细胞进入S和G2/M期,使细胞停滞于G0/G1期,与空白对照组比较,差异有统计学意义,且氧化苦参碱高剂量组最明显(P〈0.05)。5、10、50μmol/L氧化苦参碱作用细胞96h后的凋亡率分别为(10.96±0.41)%、(20.74±1.09)%、(27.05±2.00)%,与对照组(4.66±1.26)%比较,差异均有统计学意义(均P〈0.05)。氧化苦参碱浓度越高,抑制作用越明显,呈明显的剂量依赖效应。50¨mol/L氧化苦参碱作用于细胞48h、72h、96h后的凋亡率分别为(6.65±1.49)%、(14.82±5.79)%、(27.05±2.00)%,与对照组各时间点(4.01±0.86)%、(4.31±0.68)%、(4.66±1.26)%比较,差异均有统计学意义(均P〈0.05)。氧化苦参碱作用时间越长,抑制作用也越明显,呈明显的时间依赖效应。结论氧化苦参碱可通过时间依赖性和剂量依赖性方式,促使细胞G2/M期阻滞和抑制细胞有丝分裂,从而诱导人体骨肉瘤细胞凋亡。
Objective To study the apoptosis and proliferation of osteosarcoma cells ( MG-63 ) with the treatment of oxymatrine (OM) . Method OM with different concentrations was added in culture medium system for various lengths of time of induction, was detected by flow cytometry in human osteosarcoma cells ( MG-63 ) cell proliferation, cell cycle and apoptosis. Results 1-50 pμmol/L matrine could inhibit oxidation of osteosarcoma cells, and block MG-63 cells, cells from entering into the S and G2/M phase, the cells were arrested in G0/G1 phase. Compared with the blank control group, there was a significant difference, with the high dose group most significant, the most significant ( P〈0.05 ) .It was also significantly different in oxymatrine plus cisplatin group and blank control group (P〈0.05) . The apoptosis rates of 5, 10, 50 μmol/L oxymatrine in cell for 96h were ( 10.96 ± 0.41 ) %, ( 20.74 ± 1.09 ) %, ( 27.05 ± 2.00 ) %, and difference was significant compared with that in the control group [ ( 2.07 ± 1.06 ) % ] ( all P〈0.05 ) .Oxymatrine higher the concentration of the more obvious inhibition, showing a clear dose-dependent effect. The apoptotic rates with 50 p~ mol/L oxymatrine in cell for 48h, 72h, 96h were ( 6.65 ± 1.49 ) %, (14.82 ±5.79)%, (27.05±2.00)%, With the control group at all time points [ (4.31±0.68)%, (4.01±0.86)%, ( 4.66 ± 1.26 ) % ] , the differences were significant ( P〈0.05 ) .Oxymatrine were more effective with longer time of inhabitatation, with an obvious time-dependent effect. Conclusion Oxymatrine can promote cell G2/M phase arrest and inhibit cell mitosis, thereby inducing apoptosis in human osteosarcoma cells with time-dependent and dose-dependent manner.
