左归丸加黄芪对S180小鼠化疗后卵巢功能损害和抑瘤作用的研究 被引量:3

摘要 目的探讨左归丸加黄芪在干预环磷酰胺(CTX)联合顺铂(DDP)诱导的S180小鼠生殖内分泌功能损害的基础上对S180小鼠皮下肿瘤生长的影响。方法将80只C57BL/6雌性小鼠随机选取14只作为空白对照组,余皮下接种S180肿瘤细胞后随机分为3组,即模型对照组、化疗组、中药+化疗组,每组22只。动态观察肿瘤生长及动情周期,于用药前后分批处死小鼠,称取体质量、卵巢质量、肿瘤质量计算抑瘤率;采用酶联免疫吸附(ELISA)法动态检测各组小鼠血清的卵泡刺激素(FSH)、黄体生成素(LH)及雌二醇(E2)水平。每组剩余小鼠按2∶1分别与成年雄性小鼠合笼,观察生育情况。结果①给药前各组血清FSH、LH、E2水平无显著性差异(P均>0.05);给药第11天及第26天,化疗组FSH、E2水平与各组比较均有显著性差异(P均<0.01),中药+化疗组与两对照组比较无明显性差异(P均>0.05)。LH水平化疗组与中药+化疗组比较无显著性差异(P>0.05);但化疗组与两对照组比较均有显著性差异(P均<0.01)。给药第26天,化疗组FSH、LH水平与各组比较均有显著性差异(P均<0.01),中药+化疗组与两对照组比较无显著性差异(P均>0.05)。化疗组和中药+化疗组E2水平无明显差异(P>0.05);化疗组明显低于两对照组(P<0.01)。②模型对照组肿瘤质量均明显高于化疗组和中药+化疗组(P均<0.01);化疗组和中药+化疗组间比较无显著性差异(P>0.05);化疗组和中药+化疗组抑瘤率均大于60%。结论左归丸加黄芪对化疗后小鼠卵巢生殖内分泌功能的损害有保护作用;并可增强化疗对肿瘤的抑制作用。 Objective It is to investigate the effects of Zuoguiwan plus HuangQi on cyclophosphamid(CTX) combined with cisplatin(DDP)-induce ovarian reproductive endocrinology function damage in mice inoculate with SlgO murine sarcoma cells. Methods Eighty C57BL/6 female mice were divided randomly into four groups: normal control group, model control group, CTX + DDP group, herbs + CTX + DDP group. Levels of serum follicle-stimulating hormone (FSH) , luteinizing hor- mone(LH) and estradiol( E2 ) were measured successively by the enzyme-linked immunosorbent assay (ELISA) method, and mice of groups were killed before medication, in the eleventh day, in the twenty-sixth day of medication, respectively to compare weight of the tumor and efficacity of antitumor, the weight of the ovaries and mice. Results The levels of serum FSH, E2, LH in the four groups were not different significantly than the others before medication (P 〉 0.05 ). In the eleventh day the levels of serum FSH, E2 in the CTX + 1)DP group was higher significantly than the others( P 〈0.01 ) , but the difference between the herbs + CTX + DDP group between the two control groups was not significant (P 〉 0.05 ). The result of LH was similar in CTX + DDP group and herbs + CTX + DDP group, but that in the herbs + CTX + DDP group was significantly higher than that in the two control groups (P 〉 0.05). In the twenty-sixth day the levels of serum FSH, LH in the CTX + DDP group was higher sig- nificantly than that of the others (P 〈0.01 ) , but that in the herbs + CTX + DDP group was not significant different than the two control groups (P 〉 0.05 ). There was no difference in E2 belween groups of CTX + DDP and herbs + CTX + DDP, but that in CTX + DDP group was obviouly lower than that in the two control groups. The weight of tumors in the groups of CTX + DDP and herbs + CTX + DDP were lower highly than that of the control groups ( P 〈 O. 01 ) , but the former two groups had no significant difference. The inhibition rate in the groups of CTX + DDP and herbs + CTX + DDP was higher than 60%. Conclu-sion Zuoguiwan plus Huangqi could reduce the chemotherapy-induce ovarian function damage and enhance the antitumor effect of chemotherapy drugs.
