奥沙利铂与其他抗癌药物的联用,可以提高疗效及减少因剂量过大导致的不良反应,分子靶向药物也陆续应用于抗肿瘤治疗,这对于实现肿瘤患者个体化治疗具有重要意义。在此从药动学特点、作用机制、耐药机制、不良反应及临床应用等方面综合论述新一代铂类抗癌药物奥沙利铂的研究进展,为奥沙利铂剂量个体化治疗提供一定的参考;从细胞周期来阐述其耐药机制,总结传统耐药机制的理论。

Oxaliplatin in combination with other anti-neoplastic drugs can enhance the curative effect and reduce the side effects induced by excessive doses.Molecularly targeted agents are also successively used in antineoplastic therapy,which has great significance for individual therapy.Here is to make a review on the research progress of a new generation of anticancer drug oxaliplatin from the perspectives of pharmacokinetics characteristics,mechanism of action,mechanism of resistance,adverse reactions and clinical application etc.,to provide a certain reference for the individualized oxaliplatin dosage;and elaborate the drug resistance mechanism from the perspective of cell cycles,and make a summary of the conventional drug-resistance mechanism theories.
