摘要
2型糖尿病(T2DM)是一种多基因遗传病,多种基因和环境因素共同参与T2DM的发病。免疫在胰岛素作用、血糖稳态和T2DM中发挥重要作用。固有免疫反应是连接外界环境与机体相互作用的重要系统,固有免疫相关炎性途径的基因突变可能影响到T2DM危险因素的代谢表型,参与疾病的发病。Toll样受体4(TLR4)是固有免疫系统的核心介质,在T2DM中起着重要的作用。TLR4基因Asp299Gly及Thr399Ile的突变,可影响TLR4蛋白的结构,引起感染风险增加的同时也会降低慢性炎症相关疾病(如T2DM)的患病风险。
Type 2 diabetes mellitus(T2DM)is a disease of multifactorial inheritance.Multiple of genetic and environmental factors participate the development of the disease.Immune system plays an important role in insulin action,glucose homeostasis and T2DM.Inherent immunity is an important mechanism connecting the environment and human body.The genetic mutations of innate immune related inflammatory pathways may affect the metabolic phenotypes of T2DM risk factors and participate the process of the disease.Toll-like receptor 4(TRL4),a central mediator of the innate immune response,plays an important role in T2DM.TLR4 Asp299Gly and Thr399Ile polymorphisms result in the changes of TLR4 protein and are associated with the increased prevalence of infectious diseases and the decreased prevalence of chronic inflammatory diseases such as T2DM.
