摘要 通过染色法显示DNA是高校生物类专业常做实验之一,Feulgen染色是显示DNA的传统染色方法。在教学实践中由于取材不同,DNA的染色效果表现出较大差异。本研究以洋葱、玉米、蚕豆、豌豆为材料,对其不同组织进行Feulgen染色,通过比较染色效果及操作方法的难易,选择出最适合用Feulgen染色法显示DNA的实验材料。结果表明:(1)从观察结果来看,洋葱内表皮和洋葱根尖的染色效果最好;(2)从实验的操作过程来看,洋葱内表皮的取材及操作方法是最简单的一种。所以洋葱内表皮最适合用于实验教学中Feulgen染色法显示DNA。 Displaying DNA through staining method is one of required biology experiments in most universities. Feulgen is the traditional staining method to display DNA. In teaching practice, DNA staining results show large differences according to different experiment materials. In this study, onion, corn, bean and pea were used as materials and Feulgen staining method was applied to stain different tissues of the above materials. By comparing staining results and the difficulty of the op- eration, the most suitable experimental material would be selected for DNA Feulgen staining method. The results showed that : ( 1 ) from the results of observation, the staining effects of onion epidermal and root tip were the best ; ( 2 ) from the ex- perimental operation process, the operation of onion epidermal was the simplest. Comprehensively considering, onion skin is the most suitable material for Feulgen staining method to display DNA.
