摘要
目的:探讨体外培养来自硬皮病患者的成纤维细胞中转化生长因子(TGF)β受体活化的Smad3及Smad7 mRNA表达水平。方法:体外培养4例硬皮病患者(患者组)及4名正常对照者(对照组)组织的成纤维细胞,采用反转录聚合酶链反应(RT-PCR)方法,分别检测硬皮病患者组成纤维细胞及正常对照组成纤维细胞中Smad3和Smad7 mRNA的表达情况,并比较两组间的表达差异。结果:体外培养的硬皮病患者组成纤维细胞的Smad3及Smad7 mRNA表达水平均要显著高于对照组的成纤维细胞(P<0.05)。结论:Smad3在硬皮病中的上调,提示Smad3在硬皮病的发病过程中可能与TGF-β协同作用,共同促进了硬皮病的纤维化过程。而Smad7在硬皮病中的表达升高,可能是机体的一种负反馈调节作用。
Objective: To explore the expression levels of TGF-13 receptor-activated Smad3 and Smad7 in cultured scleroderma fibroblasts. Methods: The expression levels of Smad3 and Smad7 mRNA were detected by reverse transcription polymerase chain reaction(RT-PCR) in cultured fibroblasts from 4 normal controls and 4 scleroderma patients, and the levels between the two groups were compared. Results: The expression levels of Smad3 and Smad7 mRNA in cultured scleroderma fibroblasts were significantly up-regulated in comparison with those in cultured fibroblasts of normal control (P〈 0.05). Conclusions: Up-regulation of Smad3 in cultured scleroderma fibroblasts may be synergetie with TGF-13 and jointly promote the process of fibrosis in scleroderma. The elevated expression of Smad7 may play a negative feedback regulation in the pathogenesis of seleroderma.
