日本血吸虫感染者尿液诊断标志物的电泳筛选及其单克隆抗体的制备和初步应用 被引量:5

摘要 目的 筛选日本血吸虫感染者尿液检测探针。方法 采用电泳方法从日本血吸虫感染者尿液筛选诊断标志物,以此为抗原制备McAb,并用ELISA对尿样进行了初步检测。结果 PAGE电泳法筛选到一条分子量约30kDa 的糖蛋白带;以此糖蛋白为抗原免疫BALB/c 小鼠制备单克隆抗体,获得2 株McAb - NP56 和NP54;用NP56 以间接ELISA法检测10 份日本血吸虫感染者尿液(EPG中位数为69),浓缩50 倍尿液和原尿分别有9 份和5 份呈阳性。结论 本研究制备的McAb- NP56 有望作为日本血吸虫慢性病人和轻度感染者尿液的检测探针应用于现场。 Aim Screening detection probes of the urine of individuals infected with S japonicum Methods Three Kind of electrophoreses were used to screen diagnostic marker from the urine of individuals infected with S japonicum,hybrids technic was performed to produce McAbs used the diagnostic marker as antigen and the urine samples were detected by ELISA with one of the produced McAbs Results As diagnostic marker,a band of glycoprotein with molecular weight 30kDa was screened with PAGE Two lines of McAbs,named NP56 and NP54,were obtained 10 concentrated and 10 original urine samples were detected by NP56 with ELISA It showed that 9 and 5 samples were positive respectively Conclusion The results above indicate that the McAb NP56 is of potential value in the diagnoses for the chronic cases and individuals with 1ight infection with S japonicum
