小鼠增龄过程中胸腺细胞的退行性改变

摘要 目的:探讨在小鼠增龄过程中胸腺细胞退行性改变的机制。方法:选用20只不同月龄的昆明小鼠,共分为4组(乳鼠组、1月龄鼠组、3月龄鼠组及6月龄鼠组),用颈椎脱臼法处死小鼠,取出胸腺,通过HE染色检测各月龄小鼠胸腺细胞的状态和分布变化;采用流式细胞仪检测各月龄小鼠胸腺细胞的自然凋亡率及细胞周期各时相的细胞比例。结果:6月龄鼠组小鼠胸腺严重萎缩,胸腺细胞自然凋亡率与其他组比较,差异具有统计学意义。各组小鼠不同细胞周期的胰腺细胞分布不同,S期细胞凋亡速度最快。结论:小鼠增龄相关的退行性改变可能主要作用于S期进入G_2期的相关蛋白。 Objective:To explore the mechanism of degenerative changes in mice thymocytes during aging process. Methods: Twenty Kunming mice at different months of age were selected and divided into 4 groups (suckling mices, 1-month-old, 3-month-old and 6-month-old mice). Then the mice were sacrificed by cervical dislocation and extracted the thymus. The state and distribution changes were detected in mice of different months by HE staining, and apoptosis rate and cell proportion of difference phases by flow cytometry. Results: serious atrophy was seen in thymus in 6-month-old mice whose apoptosis rate was different from control group. The distribution of pancreas cells was different in different cell phases. Notably, the apoptosis rate was the fastest in S phase. Conclusion: The age-related degenerative changes might mainly affect the related proteins from S phase to G2 phase in mice.
