摘要
目的 探讨三氧化二砷 (As2 O3)对人肝癌细胞系 Hep G2的生长及核基质蛋白的影响。方法 应用生长指数分析和高分辨 SDS-聚丙烯酰胺凝胶双向电泳比较用 As2 O3处理后的 Hep G2细胞生长及核基质蛋白的改变。结果 As2 O3抑制Hep G2的生长 ,双向电泳显示经 As2 O3处理后 Hep G2细胞内出现新的核基质蛋白 ,并且该蛋白是细胞特异性的。结论 核基质蛋白是某些抗癌药物的作用靶子 ,As2 O3抑制肝癌细胞的生长可望用于临床治疗。
Objective To investigate the effect of arsenic trioxide (As 2O 3)on growth and nuclear matrix proteins of human hepatoblastoma cell line HepG2.Methods The growth rate of HepG2 treated with As 2O 3 was studied.By using high resolution two dimensional polyacrylamide gel electrophoresis,the nuclear matrix proteins of treated or untreated HepG2 were compared.Results The growth rate of HepG2 treated with As 2O 3 was reduced.2 D gel analysis showed the presence of novel nuclear matrix proteins in As 2O 3 treated tumor cells and these proteins are cell line specific.Conclusion As 2O 3 may reduce the growth of HepG2 cells.Nuclear matrix protein may be a target of As 2O 3.
