HBV DNA聚合酶在评价抗HBV药物中的应用 被引量:4

摘要 本文应用微量直接法检测血清中 HBV DNAP并对海洋药物“911”抗 HBV作用进行了评价 ,结果表明 :PFA对DNAP有较强的抑制作用 ,5 0μg/ ml剂量能全部阻断 DNAP的活性 ,1.2 5μg/ ml能抑制 5 .6 %的 DNAP活性 ,与有关报道一致。“911”对 HBV DNAP有一定的抑制作用 ,经两次平行样本重复实验 ,其结果基本一致。其平均半数抑制剂量为 4.15 mg/ ml。证明微量直接法体外测定药物对 HBV DNAP的抑制作用 ,是评价抗 HBV药物的一种快速简便的方法。 ?is a new medicine from sea plant(sea weed).To determine the inhibitory activity against HBV DNA polymerase in vitro,HBV DNA polymerase activity was detected by the method of Qimin Tao et al with a few modifications,“911” was added to the experimental samples with the concentration at 100,50,25,12 5,6 25,3 125,1 075 and 0 5mg/ml.PFA as a positive control medicine was added to the positive samples at different dose.The results showed that the inhibitory rate of PFA against DNA polymerase was 100% at 50μg/ml and 5 6% at 1 25μg/ml.The inhibitory rate of “911” against DNA polymerase was 90 8% at 100mg/ml and 4 1% at 0 5mg/ml.The average 50% inhibitory concentration(IC 50 ) was 4 15mg/ml.The results suggest that detection of HBV DNA polymerase activity in vitro is a simple and fast method to evaluate the effect of anti HBV medicine.
