干细胞因子SOX2调控Wnt/β-Catenin信号通路在脑胶质母细胞瘤发生中的机制研究 被引量:1

摘要 目的通过检测脑胶质母细胞瘤(GBM)组织中干细胞因子SOX2及β-Catenin蛋白的表达,探讨SOX2及Wnt/β-Catenin信号通路在GBM发生中的作用机制。方法脑立体定向仪定向注射C6细胞至70只SD大鼠脑尾状核建立GBM动物模型,处死大鼠取脑肿瘤组织分离、鉴定GBM干细胞将其传代培养,20只正常SD大鼠脑组织作为对照,应用SP免疫法检测模型组GBM干细胞组织和正常组脑组织大鼠SOX2、β-Catenin蛋白的表达。结果 GBM干细胞组织中SOX2和β-Catenin的表达均显著高于正常脑组织(P<0.01),二者在GBM干细胞中的表达呈正相关(r=0.372,P<0.05)。结论干细胞因子SOX2可能通过调控Wnt/β-Catenin信号通路,在GBM的发生发展过程中发挥重要作用,为针对GBM干细胞治疗该病提供了新靶点。 Objective To investigate the mechanism of SOX2 and Wnt/β-Catenin pathway in Glioblastoma Multiforme's occurrence by observing the expression of stem cell factor SOX2 and protein level of β-Catenin. Methods C6 cells were implanted in brains of 70 rats by using stereo-direction to set up GBM rat model. Then the rats were sacrificed. GBM stem cells in brain tumor tissue were isolated,identified and subcultured. The 20 normal SD rat brain tissue were used as controls. Immunohistochemistry analysis was used to detect the level of SOX2 and β-Catenin expression. Results The expression of SOX2 and β-Catenin in Glioblastoma Multiformestem cells were both higher than that of normal brain tissues(P〈0.05). There had a positive correlation between Notch1 and Bmil expressionl r = 0. 372,P〈0.05 ). Conclusion Stem cell factor SOX2 may play an impaortant role in ocurrence and development of Glioblastoma Multiforme by regulatory of Wnt/β-Catenin pathway. It provides a new target for the GBM stem cells in the treatment of Glioblastoma Multiforme.
